大鼠活体肺功能检测方法的建立及环境因素与 体质量对其的影响

Establishment of a Method for Pulmonary Function Test System in Rats and Influence of Environmental Factors and Body Weight

摘要: 目的 建立一种稳定可靠、成功率高的大鼠活体肺功能检测方法。 方法 校准小动物活体肺功能检测系统 后,大鼠气管插管,检测肺功能各项指标,观察检测过程中外界环境变化、大鼠体质量改变对肺功能参数的影响。 结果 外界的气流变化、震动因素会影响压力传感器,引起波形波动;体质量较大的大鼠( 340 ~ 370) g 功能残气量 ( FRC) 、动态肺 顺 应 性 ( Cdyn) 、第 100 毫 秒 用 力 呼 气 容 积 ( FEV100) 、 用 力 肺 活 量 ( FVC) 、 最 大 呼 气 中 期 流 量 (MMEF)均显著高于体质量较小大鼠(280 ~ 300) g,体质量对吸气阻力( RI) 没有明显影响。 结论 成功建立了简 便可靠的大鼠活体肺功能检测方法;大鼠活体肺功能检测中要确保环境的稳定以及大鼠体质量的一致性。

Abstract: Objective To develop a stable and reliable method for pulmonary function testing in rats. Method Before the experiments, the small animal pulmonary function test system was calibrated. Lung function indices of rats were measured by the system after tracheal intubation. The influence of environmental factors and body weight on lung function indices was evaluated. Result Environmental factors, including changes of air pressure or airflow, can influence the pressure sensor and alter the waveform. The rats with heavier body weights (340-370) g showed significantly increased functional residual capacity ( FRC ) , dynamic compliance ( Cdyn ) , inspiratory resistance (RI) , forced expiratory volume in 100 ms ( FEV100) , forced vital capacity ( FVC) and mean mid expiratory flow (MMEF) when compared to the rats with lower body weights ( 280 - 300) g. However, the body weights of rats did not show significant effects on inspiratory resistance ( RI) . Conclusion An easy and reliable method was successfully established for pulmonary function testing in rats. This experiment should be performed in a stable environment, and the body weights between different groups should be consistent.
